Gene editing

Achieve more efficient and specific genome editing with ssDNA and oeDNA® templates, designed to enable precise insertion via homology-directed repair (HDR). High quality double and single stranded DNA templates are synthesized by our enzymatic, scalable method using our proprietary high-fidelity enzymes to support cell and gene therapy projects.

Free from bacterial sequences, including antibiotic resistance genes, linear synthetic DNA templates improve safety and compliance for use in in vitro, ex vivo, and in vivo applications. Our synthetic DNA platform can be custom designed with varying lengths of homology arms and is suitable for complex sequences, such as high GC sequences.

ssDNA

ssDNA - Single-Stranded DNA - a linear, single stranded DNA product where both 3’ and 5’ ends are stabilized. Ideal for genome editing by (HDR), offering improved cytotoxicity profile.

Benefits of using ssDNA

  • Production of long ssDNA, up to 10 kb

  • Ability to manufacture batches up to 1 mg of ssDNA

  • Lower toxicity compared to dsDNA templates

  • The ends of the ssDNA construct are protected from exonucleases

  • No bacterial sequences or antibiotic resistance genes

oeDNA®

oeDNA® is a linear open-ended, double stranded DNA product where both 3’ and 5’ ends are open.  Constructs can be generated with blunt ends, overhangs, or a combination of both.

Benefits of oeDNA®

  • Enhanced sequence integrity

  • Production of sequences from 140 bp up to 20 kb

  • No bacterial sequences or antibiotic resistance genes

  • More suitable for scale up, batches up to 10 g

  • Available in RUO, HQ and GMP grade
